体外培养人外周血白细胞中阿德福韦酯诱生干扰素的作用研究

Objective To observe whether adefovir dipivoxil has antiviral immunological effect in vitro culturing white blood cells of patients with chronic hepatitis B (CHB) and normal people by determining the IFN activity in culture medium.Methods The blood specimen were gathered from twenty CHB patients and twenty health people, then WBC were cultured in medium containing adefovir dipivoxil (5ug/ml, 7.5ug/ml and 10ug/ml) and control group with no adefovir dipivoxil.At different time (24h、48h、72h) after agent added, culture medium were harvested.Meanwhile, IFN activity of each sample was determined by cytopathic effect inhibition assay.Results At different time after drugs were added, in health people, the titers of IFN activity in drug groups (5ug/ml, 7.5ug/ml and 10ug/ml) improved significantly at 24h (P<0.001) , in 5ug/ml group it was significantly higher at 72h hours than that at 24h (P=0.004) ;there were no significant difference between drug groups and control groups in CHB patients.Conclusion Adefovir dipivoxil can induce WBC of healthy people to secrete interferon to certain extent, but this function isn't significant in WBC of patients with CHB.
