氧化苦参碱对肿瘤坏死因子诱导的人L02肝细胞凋亡的影响

we investigate the effects of Oxy on apoptosis induced by rhTNF α in human liver cell line L02.Cell viability and proliferation was assayed by trypan blue stain and H 3 TdR co operalion assay.Flowcytometer was used for the cell cycle analysis using propidium iodide DNA binding.Apoptosis can be induced by rhTNF α in L02 cells.Evaluation of the PI DNA binding by flow cytometry showed abolition of G1, S, and G2/M phases and their replacement by a sub 2N AP peak of fragmented DNA.Characteristic apoptic condensations, ranging from single ball condensations to budding, and apoptic bodies were found in cells.Cells treated with Oxy+rhTNF α 15ng/ml partly retain the ability of proliferation, showing no significantly sub 2N Ap peak of fragmented DNA by flow cytometry.Oxy may have inhibitory activity on apoptosis of L02 cells induced by rhTNF α.
