Research advances in genetic susceptibility to non- alcoholic fatty liver disease

摘要: 非酒精性脂肪性肝病(NAFLD)是临床上最为常见的肝病之一,其病因及发病机制较为复杂。作为一种遗传-环境-代谢-应激相关性疾病,遗传因素在NAFLD的发病中起重要作用。为了认识NAFLD的发病机制中遗传因素的作用,探求NAFLD新的临床治疗靶点,归纳和整理了近年来与NAFLD相关的遗传易感基因,特别是对通过全基因组关联分析发现的基因,包括含patatin样磷酯酶域3、瘦素受体、脂联素、肿瘤坏死因子α等进行了论述。Abstract: Non- alcoholic fatty liver disease( NAFLD) is one of the most common liver diseases,with complicated etiology and pathogenesis. As a heredity- environment- metabolism- stress disease,the genetic factors play an important role in the pathogenesis of NAFLD. In order to identify the role of genetic factors in the pathogenesis of NAFLD and to find out new clinical therapeutic targets,the present review summarizes the susceptibility genes of NAFLD,especially those identified by genome- wide association study,including the genes of patatin- like phospholipase domain- containing 3,leptin receptor,adiponectin,and tumor necrosis factor- ɑ.
