Effect of Ganfukang on JAK2 and STAT3 expression in rats with liver fibrosis

摘要: 目的观察中药肝复康对肝纤维化大鼠肝组织Janus激酶2(JAK2)及信号转导子和转录激活子3(STAT3)表达的影响,并对其在JAK2/STAT3信号通路上治疗肝纤维化的作用机制进行初步探讨。方法采用10%四氯化碳皮下注射制备肝纤维化模型,于造模第9周给予肝复康治疗12周。通过测定血清中ALT、AST活性及白蛋白和总蛋白的含量来反映肝脏功能,HE染色法观察肝组织病理学变化,RT-PCR观察JAK2和STAT3 mR-NA的表达。结果经肝复康治疗后,中剂量治疗组与模型组相比较,肝脏的组织学和血清学指标均明显改善,肝组织JAK2、STAT3 mRNA的表达显著减少(P<0.01)。结论 (1)JAK2/STAT3信号通路在肝纤维化的形成过程中起重要作用;(2)肝复康对肝纤维化有疗效,其作用机制可能与降低肝组织JAK2和STAT3的表达,阻断JAK2/STAT3信号通路有关。Abstract: Objective To study the effect of traditional Chinese medicine-Ganfukang on the expression of Janus kinase 2 (JAK2) and signal transducer and activator of transcription 3 (STAT3) in rat with liver fibrosis, and explore its mecha-nism on JAK2 /STAT3 pathway.Methods The hepatic fibrosis in rats was induced by CCl4, and then the rats were treated with Ganfukang for 13 weeks.The serum levels of ALT, AST, albumin and total protein were measured.The pathological characteristics of hepatic tissues were detected by HE staining.Hepatic JAK2 and STAT3 expressions were detected by RT-PCR.Results Compared to the control group, histological and serum indexes were improved in the middle dose group.The expression of JAK2 and STAT3 mRNA in the middle dose group decreased significantly compared to the control group (P <0.01) .Conclusion JAK2/STAT3 signal transduction pathway plays an important role in the process of hepatic fibro-sis.Ganfukang had a therapeutic effect on hepatic fibrosis.The mechanism of action may be related to the decreased ex-pression of JAK2 and STAT3 of hepatic tissue and blocking of JAK2 /STAT3 signal transduction pathway.
