Effect of baicalin of the expression of NF-κB in acute liver failure in rats

摘要: 目的观察急性肝衰竭大鼠肝脏NF-κB的表达水平及黄芩苷对其表达的影响。方法雄性SD大鼠106只随机分为正常组、肝衰竭组和黄芩苷组。肝衰竭组和黄芩苷组采用腹腔注射D-氨基半乳糖制备大鼠急性肝衰竭模型;黄芩苷组于造模后每隔12 h以120 mg/kg剂量腹腔注射。造模后24 h、72 h、120 h和168 h处死大鼠。全自动生化仪检测血清ALT、AST和TBil水平;HE染色观察肝脏病理学变化;RT-PCR法检测肝组织NF-κB、TNFα、Caspase-3 mRNA表达;免疫组化法检测肝组织NF-κB蛋白表达。结果黄芩苷组大鼠168 h存活率明显高于肝衰竭组。黄芩苷组大鼠各时间点血清ALT、AST、TBil水平较肝衰竭组明显降低(F=173.584,158.329,74.902;P<0.01)。黄芩苷组NF-κB、TNFα、Caspase-3 mRNA表达趋势与肝衰竭组相同,72 h达高峰,但表达量较肝衰竭组明显减少,差异有统计学意义(F=603.801,42.174,27.222,P<0.001,P<0.001,P=0.001)。黄芩苷组NF-κB蛋白的表达也于72...Abstract: Objective To observe the expression of NF-κB in acute liver failure in rats and the effect of baicalin on the expression of NF-κB.Methods 106 male SD rats were randomly divided into normal group, model group and baicalin group.The model group and baicalin group produced the animal model of acute liver failure by intraperitoneal injection with D-galactosamine (D-GalN) .Baicalin (120 mg/kg) was treated intraperitoneally (i.p.) at every 12 h after D-GalN was injected in rats.We chose 6 rats randomly at the time of 24, 72, 120 and 168 h from model group and baicalin group for detecting the indexes we needed.The levels of ALT、AST and TBil in serum were detected with automatic biochemical analyzer.Sections of liver tissue were stained with hematoxylin-eosin and were observed under optical microscope.RT-PCR (reverse transcription-polymerase chain reaction) was used to study the expression of NF-κB, TNF α and caspase-3 mRNA in the liver.NF-κB in rat liver was determined at different time points with immunohistochemical method.Results The 168 hours survival rate of therapy group was significantly higher than the model group (P<0.05) .Compared with model group, the levels of ALT、AST and TBil of baicalin group were significantly reduced, the discrepancy between model group and baicalin group had statistical significance (P<0.01 or 0.05) .Compared with model group, the tendency of NF-κB, TNF α and Caspase-3 mRNA expression in baicalin group was in accordance with the model group, which reached a peak at 72 h, the expressions were significantly less than model group, and the discrepancy between model group and baicalin group had statistical significance (P<0.001, P<0.001, P=0.001) .The tendency of NF-κB protein expression in baicalin group was in accordance with the model group, the expressions at 72 hours were significantly less than model group, and the discrepancy between model group and baicalin group had statistical significance (P=0.017) .Conclusion Baicalin can protect rats against acute liver failure by down-regulating NF-κB, TNF α and Caspase-3 expression in liver tissue and reducing of pro-inflammatory cytokines.
