摘要: 目的探讨HBVC基因型较B基因型易引起肝炎慢性化的机制。方法从GenBank分别下载C基因型和B基因型小S蛋白的氨基酸序列(即HBsAg),用CLustalW2.0生物软件分别比较C基因型和B基因型小S蛋白氨基酸序列的相似性;用神经网络的方法预测同基因型不同的小S蛋白氨基酸序列之间的T细胞表位的差异。结果通过分别比较C基因型和B基因型小S蛋白的氨基酸序列的相似性,C基因型小S蛋白的氨基酸序列之间的相似性为(90~100)%,B基因型小S蛋白的氨基酸序列之间的相似性为(94~100)%;同基因型不同的小S蛋白氨基酸序列之间的T细胞表位存在明显差异。结论C基因型小S蛋白的氨基酸序列之间较大的变异和同基因型不同的小S蛋白氨基酸序列之间的T细胞表位的差异可能是HBVC基因型较B基因型易引起肝炎慢性化的原因或部分原因。Abstract: Objective To discuss the mechanism of C genotype HBV bing easier to arose chronic hepatitis than B genotype HBV.Methods Download amino acid sequences of HBV C genotype small S protein and HBV B genotype small S protein and analyze comparability of these amino acid sequences by software CLustalW 2.0.To forecast the differerce of T cell epitope of different small S protein sequences of C genotype HBV using NN way.Results The result of comparability of small S protein of B genotype HBV and C genotype HBV showed that comparability of amino acid sequence of small S protein of B genotype HBV is 94~100%, and C genotype HBV is 90~100%;the result that genotype is same but small S protein sequence is different results in different T cell epitope.Conclusion The cause or part cause of C genotype HBV bing easier to arose chronic hepatitis than B genotype HBV can be that variation of small S protein sequence of C genotype HBV is bigger than B genotype, the bigger variation result in higher variation ratio of T cell epitope.
