The effect of Interferon treatment on cellular immuneresponses in patients with chronic hepatitis B

摘要: 目的探讨Th1/Th2类细胞因子在CHB患者PBMC表达,及干扰素治疗慢性乙型肝炎（CHB）对患者Th1/Th2类细胞因子的影响。方法30例CHB患者给予干扰素治疗,治疗前后采用双抗体夹心法检测外周血单个核细胞IL-4、IL-10和IFN-γ的水平,同时选择15例健康人群作对照组。结果CHB组PBMC培养上清分泌IL-4水平显著高于健康对照组（P<0.05）,而IFN-γ水平则显著低于健康对照组（P<0.01）;HBV DNA水平与IL-4水平呈正相关,与IFN-γ水平呈负相关。治疗结束后IL-4、IL-10水平较治疗前显著降低（P<0.01,P<0.05）,IFN-γ水平较治疗前显著升高（P<0.01）。结论CHB存在异常细胞免疫应答,干扰素治疗CHB可提高患者Th1型细胞因子水平、降低Th2型细胞因子的水平。

Abstract: Objective To explore the levels of IL-4、IL-10 and IFN-γ in culture superatants of peripheral blood mononuclear cells (PBMC) in patients with chronic hepatitis B (CHB) , and to detect the effect of interferon treatment.Methods The productions of IL-4、IL-10 and IFN-γ in culture supernatants of PBMC from 30 patients with CHB were detected pre-and post-interferon treatment, In addition, there were 15 healthy individuals as controls at the same time.Results The level of IL-4 in the patients with CHB was significantly higher than that in the control (P<0.05) , and the level of IFN-γ was significantly lower than that in the control (P<0.01) ;The level of IL-4 was positively correlated with the level of HBV DNA, but the level of IFN-γ was negatively correlated with the level of HBV DNA;After interferon therapy, the level of IL-4 and IL-10 was obviously decreased (P<0.01, P<0.05) , the level of IFN-γ was significantly increased (P<0.01) .Conclusion There is an abnormal cell-mediated immune response in CHB patients, Interferon treatment not only promoted Th1 type cytokine-mediated immune responses but also down regulated Th2 type cytokine-mediated immune responses.
