Expression of insulin-like growth factor-binding protein-2 (IGFBP2) and matrix metalloproteinase-9 (MMP9) and its clinicopathologieal significances in benign and malignant lesions of gallbladder

摘要: 目的研究胆囊良恶性病变组织中胰岛素样生长因子结合蛋白-2(IGFBP-2)和基质金属蛋白酶-9(MMP9)表达水平及其临床病理意义。方法108例胆囊腺癌、46例癌旁组织和35例慢性胆囊炎标本石蜡包埋切片,IGFBP2和MMP9染色方法为EnVisionTM免疫组化法。结果胆囊腺癌IGFBP2和MMP9表达阳性率(54.6,59.3)及其评分(2.26±1.67,2.88±1.15)明显高于癌旁组织(28.3,1.05±1.21;32.6,1.16±1.10)、慢性胆囊炎(20.0,0.77±0.99;22.9,0.79±1.18)(P<0.01)。腺瘤癌变、肿块最大径<2cm、无淋巴结转移、未侵犯周围组织及无胆囊结石病例IGFBP2和MMP9表达阳性率及其评分明显低于中、低分化腺癌、肿块最大径≥2cm、淋巴结转移、侵犯周围组织及伴胆囊结石病例(P<0.05或P<0.01);胆囊腺癌中IGFBP2表达评分与MMP9表达评分呈密切正相关(r=0.46,P<0.01)。结论IGFBP2和MMP9表达是反映胆囊癌发生、进展、生物学行为和预后的重要生物学标记物,IG...

Abstract: Objective To study on the expressive levels of IGFBP-2 and MMP9 and detect their clinicopathological significances in benign and malignant lesions of gallbladder.Methods En VisionTM immunohistochemical method was used to detect the expressive levels of IGFBP2 and MMP9 in the routinely paraffin-embedded sections of the specimens from patients with gallbladder adenocarcinoma (n=108) and its pericancerous tissues (n=46) and chronic cholecystitis (n=35) .Results The positive rates and scores of IGFBP2 and MMP9 in gallbladder adenocarcinoma (54.6%, 2.26±1.67;59.3%, 2.88±1.15) were significantly higher than those in pericancerous tissues (28.3%, 1.05±1.21;32.6%, 1.16±1.10) and chronic cholecystitis (20.0%, 0.77±0.99;22.9%, 0.79±1.18) (P<0.01) .The positive rates and scores of IGFBP-2 and MMP9 were significantly lower in the cases of adenomataus canceraion, maximal diameter of mass<2cm, no-metastasis of lymphnode, no-invasion of regional tissues, and no-gallstone than those in the ones of moderately-or low-differentiated adenocarcinoma, maximal diameter of mass=2cm, metastasis of lymphnode, invasion of regional tissues and with-gallstone (P<0.05 or P<0.01) .The closely positive correlation was found between the expressive scores of IGFBP-2 and MMP9 in gallbladder adenocarcinoma.Conclusion The expressive levels of IGFBP-2 and MMP9 might be important biological markers for reflection the carcinogenesis, progression, biological behaviors and prognosis of gallbladder adenocarcinoma.IGFBP-2 might promote the biological synthesis of MMP9 in gallbladder adenocarcinoma tissues. -
