Experiment study of metastasis suppressor gene in primary liver cancers

摘要: 目的检测转移抑制基因Kail的蛋白表达,探讨其与原发肝癌生物学行为的关系。方法采用免疫组化SP法分别测定了53例肝癌、17例正常肝脏新鲜冰冻组织中Kail蛋白的表达。结果53例原发肝癌中19例(36%)Kail蛋白表达阳性,与正常肝脏(82%)相比,差异有统计学意义(P<0.01)。早期肝癌Kail蛋白表达率明显高于中,晚期肝癌(P<0.05)。巨块癌Kail表达率高于结节癌(P<0.01)。癌细胞分化程度Ⅰ、Ⅱ级肝癌Kail表达率明显高于晚期Ⅲ、Ⅳ级(P<0.05)。结论Kail蛋白表达下降可能预示转移,且于肝癌恶性程度有关,可成为判断预后的因素之一。Abstract: Objective To explore the expression of metastasis suppressor gene Kail in primary liver cancers and its relationship to clinicopathological feature.Methods The expression of Kail protein in fresh freezing tissures from 53 primary liver cancer patients, 17 normal were examined by immuno-histochemistry SP method.Results Kail expression was observed in 19 of 53 primary liver cancers (36%) and in 14 of 17 normal liver (82%) , the latter is higher (P<0.01) .It was significantly higher in early stage than that in late stage tumors (P<0.05) .Serous liver cancers showed significantly decreased staining of Kail when compared to macrosia cancers (P<0.01) .Furthermore, Kail expression in atypia (Ⅰ、Ⅱ) was higher than that in Ⅲ、Ⅳ (P<0.05) .Conclusion The downregulation of Kail may indicate metastasis, and relate with malignant degree of liver cancer.It can be used new prognostic marker of c primary liver ancer.
