The influence of 5-hydroxytryptamine on rat with hepatic encephalopathy.

摘要: 目的探讨五羟色胺(5-HT)在大鼠急性肝衰竭(ALF)肝性脑病(HE)发病中的影响。方法应用SD大鼠,随机分为正常对照A,观察组B、C、D,每组20只。B、C、D组分别应用5%硫代乙酰胺(TAA)500mg/kg灌胃,制作ALF HE动物模型。B组和C组分别加用塞庚啶10mg/kg、20mg/kg。各组分阶段观察5-HT,肝生化指标、血氨、内毒素检测值、脑核磁共振及脑组织病理学电镜检查。结果B、C、D组5-HT测定值明显高于正常对照组(t≥3.9,P<0.01)。B、C组死亡率低于D组(x2≥0.63,P<0.05)。D组72小时5-HT值明显高于D组48小时组(t=6.1,P<0.01)。B、C组脑组织电镜检查示神经元细胞肿胀,有髓样小体形成,D组脑组织神经元细胞固缩,染色质增生,髓鞘水肿变性明显,部分有分层断裂。结论肝衰竭肝性脑病时5-HT有异常升高。应用5-HT受体拮抗剂可见脑组织病理损伤较轻,死亡率降低。

Abstract: Objective To explore the influence of 5-hydroxytryptamine on rat with hepatic encephalopathy (HE) in stage of acute liver failures (ALF) .Methods Eighty Sprague-Dawley rats were divided into four groups randomly: Group A acted as control.Model of rat with HE were induced by 5% thioacetamide (TAA) intragastric administration in group B, C and D.Group B and C were given by 5-HT receptor blocking agent (Cyproheptadine) with 10 mg/kg and 20mg/kg respectively.Each group was detected by liver function tests, 5-HT, blood ammonia, serum endotoxin, brain magnetism resonance and histopathology.Results The level of 5-HT in group B, C and D were higher than those in normal control (t≥3.9, P<0.01) .The mortality in group B and C were both lower than that in group D (x2≥0.63, P<0.05) .The level of 5-HT at 72hour in group D was higher than that at 48-hour. (t=6.1, P<0.01) .The electron microscope showed that the neuron had swollen in groups B and C and marrow-like body had formed, whereas in group D, nerve cell were contracted and chromatin were hyperplasia, myeline sheath were edema and denaturation.Conclusion The level of 5-HT was increased obviously in rat with HE.The histological damage of brain has mitigated after administration with 5-HT receptor blocking agent and the mortality had cut down.-
