Study on the relationship of APOBEC3G mRNA expression in PBMC with HBV viral load and IFN-γ in chronic hepatitis B patients.

摘要

摘要: 目的探讨慢性乙肝患者外周血单个核细胞（PBMC）APOBEC3G mRNA的水平,以及其与IFN-γ mRNA、血清HBV病毒载量及ALT的关系。方法用实时荧光定量RT-PCR的方法检测27例慢性乙肝患者以及16名健康人PMBC中APOBEC3G和IFN-γ mRNA的水平,并分别检测每个乙肝患者血清HBV病毒载量及肝功能ALT。结果慢性乙肝患者PBMC中APOBEC3G mRNA的表达水平高于健康对照组,差异具有显著性（P<0.01）,IFN-γ mRNA无明显增高,A3G还与患者血清中的病毒载量呈正相关（r=0.73,P<0.01）,但与IFN-γ及ALT水平无关（P>0.05）。结论表达增高的APOBEC3G蛋白可能只是HBV慢性感染的伴随表现,APOBEC3G蛋白是否具有抗HBV临床应用价值,还需要更加深入的研究探讨。

Abstract: Objective To assess the level of APOBEC3G mRNA in peripheral blood monouclear cells (PMBC) from chronic hepatitis B patients and to examine the relationship of it with IFN-γ mRNA、HBVviral loads and ALT.Methods real-time fluorescence quantitative RT-PCR was used to quantify APOBEC3G and IFN-γ mRNA levels in PBMC from 16 HBV-uninfected subjects and 27 patients with chronic hepatitis B discease.Besides, HBV viral loads and ALT were detected too.Results APOBEC3G mRNA levels in PBMC of hepatitis B patients were higher than those of HBV-uninfected subjects (P<0.01) , while the levels of IFN-γ mRNA were not enhanced as compared to HBV-uninfected subjects.Futhermore, the relative level of A3G mRNA were correlative with HBV viral loads directly (r=0.73, P<0.01) , In Contrast, ALT and IFN-γ mRNA was not relative with APOBEC3G mRNA transcription in vivo.Conclusion The enhanced expression of APOBEC3G might be a concomitant phenomenon of HBV infection.It needs much further investigation to determine whether APOBEC3G proteins have clinical value against HBV infection.
