Preliminary study on proteomic patterns in hepatic tissue to identify HBV related hepatic firbrosis.

摘要: 目的获得早期诊断乙型肝炎病毒(HBV)相关肝纤维化的肝组织蛋白质组模式。方法采用双向凝胶电泳(2-DE)技术和计算机辅助的图像分析方法获得肝纤维化组织和正常肝组织的蛋白质表达谱。先对6例肝纤维化组织和正常肝组织蛋白质表达谱进行分析,在肝纤维化组织蛋白质表达谱中发现具有鉴别肝纤维化组织与正常肝组织的蛋白质组模式,再应用该模式对14例未知的肝纤维化组织和正常肝组织2-DE图谱进行检测分析。结果未知的肝纤维化组织与正常肝组织均得到准确鉴定,灵敏度和特异均为100%。结论HBV相关肝纤维化组织的蛋白质表达模式能准确判断肝纤维化,对该模式中的蛋白质点进一步分析有助于研究HBV相关肝纤维化的诊断标记。Abstract: Objective To identify proteomic patterns in hepatic tissues for diagnosing early HBV related hepatic fibrosis.Methods Proteomic spectra were generated by two-dimensional gel eletrophoresis (2-DE) .A preliminary "training" set of spectra derived from analysis of 3 hepatic fibrosis tissue and 3 healthy liver tissues, a proteomic patterns that completely discriminated cancer from non-cancer was identified.The discovered proteomic patterns then was used to classify an independent set of 14 mask samples: 10 from hepatic fibrosis tissues, and 4 from healthy liver tissues.Results The discriminatory pattern correctly identified all hepatic fibrosis tissues and healthy liver tissues in the mask set.The result yielded a sensitivity of 100%, specificity of 100%.Conclusion Apparent proteome difference is detected between hepatic fibrosis tissues and healthy liver tissues.Further analysis on the different spots of interest may be found the pathogenesis of hepatic fibrosis or early diagnosis marker of hepatic fibrosis.
