Effect of Curcumin on proliferation and extracellular matrix secretion of rat hepatic stellate cells in vitro

摘要: 观察姜黄素对大鼠肝星状细胞 (HSC)增殖及分泌细胞外基质的影响。用不同浓度的姜黄素处理大鼠肝星状细胞 ;以MTT比色法测定细胞的增殖 ;放射免疫法测定透明质酸和层粘连蛋白 ;酶联免疫吸附法 (ELISA)测定Ⅰ型胶原的水平。姜黄素可剂量依赖性地抑制HSC增殖 ,不同程度抑制Ⅰ型胶原、透明质酸和层粘连蛋白分泌。姜黄素可显著抑制HSC的增殖及分泌细胞外基质。Abstract: To investigate the effects of Curcumin on proliferation and extracellular matrix secretion of rat hepatic stellate cells in vitro. HSC-T6 were incubated with different concentration of Curcumin for 24 hours. MTT colorimetry was used for assaying proliferation of HSC. Type Ⅰ collage was determined by ELISA. Hyaluronic acid and Laminin were measured by radioimmunoassay. addition of (1～20) μmol/L curcumin into culture medium had no cytotoxicity to HSC, but could significantly inhibi HSC proliferation, and also inhibit Type Ⅰ collage, hyaluronic acid and laminin secretion in different degree. Curcumin can significantly inhibi HSC proliferation and extracellular matrix secretion.
