Significance of detecting apoptosis of PBMC and cytokine of supernatant induced by PHA in chronic hepatitis B

摘要: 探讨外周血单个核细胞凋亡和Th1/Th2型细胞因子表达在慢性乙肝发病中的作用。采用PI染色法检测单个核细胞凋亡 ,ELISA法检测IL - 2、IFN -γ、IL - 4、IL - 10等细胞因子水平。慢性乙肝患者单个核细胞凋亡明显高于对照组 (P <0 0 1) ,随着抗原刺激时间的延长细胞凋亡增加 ,有显著性意义 (P <0 0 1)。IL - 2、IFN -γ水平低于正常对照组 (P <0 0 1) ,其浓度随着培养时间的延长有轻度减低 ,但无显著意义 (P >0 0 5 )。IL - 4、IL - 10水平高于正常对照组 (P <0 0 1) ,其浓度随着培养时间的延长逐渐升高并且差异有显著意义 (P <0 0 1)。慢性乙肝患者外周血淋巴细胞凋亡增强及Th1/Th2免疫调节紊乱 ,使细胞免疫功能减弱 ,淋巴细胞凋亡和Th1/Th2的失衡在慢性乙肝发病中起一定的作用。Abstract: To investigate the significance of PBMC apoptosis and Th1/Th2 cytokine in the pathogenesis of chronic hepatitis B The apoptosis was measured by propidium iodide staining and the IL-2、IFN-γ、IL-4、IL-10 of PBMC induced by PHA was detected with the ELISA The apoptosis of PBMC was higher in hepatitis B than in control group (P<0.01) . There were significant of the increasing apoptosis in hepatitis B when the cultured time was lengthened (P<0.01) . The level of IL-2, IFN-γ was lower in hepatitis B than that in control group (P<0.01) , with the cultured time lengthening, the level of IL-2, IFN-γ slightly decreased while there were no significant (P>0.05) .The level of IL-4、IL-10 was higher in hepatitis B than that in control group (P<0.01) . And the level became higher with the cultured time lengthening, in addition there was significant (P<0.01) .The cell-mediated immunity was weakened because of the strengthened apoptosis of PBMC and imbalance of Th1/Th2 in chronic hepatitis B, lymphocyte apoptosis and imbalance of Th1/Th2 have somewhat function in the pathogenesis of chronic hepatitis B.
