Expression of PTTG and bFGF in primary gallbladder cancer and their clinical significance

摘要: 探讨垂体肿瘤转化基因 (PituitaryTumorTransformingGene ,PTTG)和碱性成纤维细胞生长因子 (basicFi broblastGrowthFactor,bFGF)在原发性胆囊癌中的表达及相互关系 ,研究它们的表达与肿瘤临床病理指标及预后的联系。应用免疫组织化学 ,检测 4 1例原发性胆囊癌及 2 0例慢性结石性胆囊炎中PTTG和bFGF的表达水平。在原发性胆囊癌中PTTG和bFGF阳性表达率分别为 85 4 %、5 6 1% ,其阳性率及表达等级均明显高于慢性胆囊炎 (P <0 0 1)。PTTG和bFGF表达等级均与胆囊癌的Nevin分期和淋巴结转移密切相关 (P <0 0 5或P <0 0 1) ;PTTG表达与bFGF表达成等级正相关 (r =0 5 2 6 ,P <0 0 1)。PTTG或bFGF阳性患者的累计生存期明显低于阴性者 (P <0 0 5 )。PTTG或bFGF与胆囊癌生物学行为及预后有密切关系 ,二者的联合检测 ,有助于原发性胆囊癌恶性程度和预后的判断

Abstract: To investigate the expression and their relation each other of PTTG (Pituitary Tumor Transforming Gene) and bFGF (basic Fibroblast Growth Factor) in primary gallbladder cancer, as well as the relationship of their expression with clinical-pathological indexes and prognosis in primary gallbladder cancer. PTTG and bFGF were examined in 41 cases of primary gallbladder cancer and 20 cases of chronic calculous cholecystitis tissues by immunohistochemical assay. The positive rate of PTTG and bFGF was 85.4% and 56.1% respectively in gallbladder cancer, whose positive expression rate and expression grade was detected singnificantly higher than that in chronic cholecystitis tissues (P<0.01) . The expression grade of PTTG and bFGF was significantly correlated to the Nevin staging and lymph node metastasis of gallbladder cancer (P<0.05 or P<0.01) :positive expression rate and expression grade of PTTG and bFGF increase with progression of Nevin stage; positive expression rate of PTTG and bFGF for patients who have lymph node metastasis was obviously higher that of no lymph node metastasis. Expression of PTTG was positively correlated with the expression of bFGF (r=0.526, P<0.01) . The PTTG or bFGF positive patients have a shorter survival time than PTTG or bFGF negative ones (P<0.05) . PTTG or bFGF is highly related to the biological behavior and prognosis of gallbladder cancer. Associated detection of PTTG and bFGF may help to judge the magligant degree and prognosis of primary gallbladder cancer.-
