Effect of Interferon-α therapy on production of IL-12 secreted by peripheral blood dendritic cells in patients with chronic hepatitis B

摘要: 探讨干扰素治疗慢性乙型肝炎 (CHB)患者对外周血树突状细胞 (DCs)分泌IL - 12的影响。采集并分离2 3例干扰素治疗前、后CHB患者的外周血单个核细胞 ,在rhIL - 4和rhGM -CSF的作用下培养 10天 ,收集上清液检测IL - 12的水平 ,以 8例健康人作为对照。干扰素治疗前患者DCs分泌IL - 12的水平显著低于对照组 (P <0 0 1) ,治疗 4个月时DCs的分泌水平较治疗前显著增加 (P <0 0 5 )。进一步分析发现 ,治疗后IL - 12的水平仅在完全应答组较治疗前显著性增加 (P <0 0 1)。提示CHBDCs分泌IL - 12的水平低下 ,干扰素治疗可以促进DCs产生IL -12 ,且其增加的程度与干扰素的疗效有关Abstract: To investigate the effect of interferon-α (IFN-α) therapy on the production of IL-12 secreted by peripheral blood dendritic cells in patients with chronic hepatitis B (CHB) . The PBMCs samples were isolated from 23 patients just before and after IFN-α treatment and cultured with rhIL-4 and rhGM-CSF for 10 days, respectively. The supernatants were collected and the levels of IL-12 were detected with ELISA. 8 healthy individuals were regarded as controls. The results showed that the level of IL-12 was significantly lower in IFN-α pretreatment group than that in control group (P<0.01) . After treatment for 4 months, the IL-12 level markedly increased compared with the in level pretreatment group (P<0.05) . Furthermore the significantly elevated level of IL-12 in posttreatment groups could only be observed in complete responder group (P<0.01) . The study indicates that DCs secrete low production of IL-12 in CHB patients. IFN-α therapy can promote DCs to secrete IL-12, and its secreting level may related to the antiviral efficacy of IFN-α.
