The protective effects of bone-marrow-derived mesenchymal stem cells on hepatic regeneration and repair of rat liver graft after reduced-size liver transplantation

  • Objective To explore the effect of bone-marrow-derived mesenchymal stem cells (BMSCs) in promoting regeneration and repair of rat liver after reduced-size liver transplantation.Methods (1) BMSCS which were taken from the bone marrow of the Wistar rats were cultured in vitro, then marked by CSFE and prepared as suspension. (2) 50% reduced-size liver transplantation models (SD donor to Wistar recipient) were established, which were divided into the experiment group (1ml BMSCs suspension was injected into portal vein) and the control group (1ml saline was injected into portal vein) .After operation, the living state of recipient was observed and the indicators about hepatic regeneration were detected through which serum and liver samples were collected and analyzed by biochemistry and immunohistochemistry respectively after transplantation.Results (1) BMSCs showed a high purity and homogeneity. (2) 2 hours after liver transplantation, all rats were recovered moving freely and drinking water. (3) There were BMSCs in the liver graft.The ratio between liver weight and body weight in the experimental group was higher than the control group.Histopathology revealed that hepatocyte injury was minor in the experimental group than the control group.Conclusion BMSCs transfused into the liver graft can promote both regeneration and repair of liver cells after reduced-size liver transplantation.
