Role of silent information regulator 1 in hepatic ischemia-reperfusion injury

  • 摘要: 肝脏缺血再灌注损伤一直是肝移植及肝脏肿瘤切除等手术中面临的难题,多种机制的参与使其变得尤为复杂,其中缺血期间的能量消耗和再灌注诱导的氧化应激是导致肝脏缺血再灌注损伤的主要机制,二者共同导致细胞死亡甚至引发肝衰竭。沉默信息调节因子(Sirt)是一类具有烟酰胺腺嘌呤二核苷酸依赖性的脱乙酰化酶,可使组蛋白及非组蛋白等转录因子脱乙酰化,在细胞凋亡、炎症反应、能量平衡及氧化应激等方面发挥重要的调节作用,哺乳动物体内沉默信息调节因子有7种,包括Sirt1~Sirt7,其中,Sirt1可通过多种信号通路来减轻肝细胞应激并调控细胞代谢途径,从而达到减轻肝脏缺血再灌注损伤程度的目的,主要对Sirt1相关信号通路在肝脏缺血再灌注损伤中的作用进行了综述。
