The expression of suppressor of cytokine signaling-1 mRNA in the peripheral blood mononuclear cells of patients with chronic hepatitis B infection

摘要: 目的检测慢性乙型肝炎（CHB）患者外周血单个核细胞（PBMCs）中,细胞信号转导抑制因子-1（SOCS-1）mRNA的表达水平,分析其表达与肝脏不同炎症活动程度之间的关系。方法 CHB患者分为轻、中、重度组各15例,乙型肝炎病毒（HBV）携带者15例为携带组,15例健康体检者作为对照组。采集外周静脉血,密度梯度离心法分离PBMCs。RT-PCR检测SOCS-1 mRNA的表达。结果重度组SOCS-1 mRNA的表达（1.15±0.12）明显高于对照组（0.86±0.10）、携带组（0.89±0.18）和轻度组（0.93±0.12）（q=4.183～5.472,P<0.01）。CHB患者SOCS-1 mRNA的表达水平与肝脏不同炎症活动程度呈正相关（rs=0.664,P<0.01）。结论 CHB患者外周血PMBCs中SOCS-1 mRNA的表达与肝脏炎症活动程度有关。

Abstract: Objective To investigate the expression of suppressor of cytokine signaling-1 (SOCS-1) in the peripheral blood mononuclear cells (PBMCs) of patients with chronic hepatitis B (CHB) , and analyze the relationship between the expression and the degree of liver inflammation.Methods CHB patients were divided into three groups: mild group, moderate group and severe group respectively with 15 cases, 15cases of HBV carriers as carry group and 15 cases of healthy volunteers as control group.The PBMCs were separated by density gradient centrifugation and the lever of SOCS-1mRNA was determined by RT-PCR.Results The lever of SOCS-1 mRNA in the severe group (1.15±0.12) was significantly higher than that in control group (0.86±0.10) , carry group (0.89±0.18) and mild group (0.93±0.12) (q=4.183～5.472, P<0.01) .The expression of SOCS-1mRNA and the level of inflammation of the liver in CHB patients were positively correlated (rs=0.664, P=0.003) .Conclusion The lever of SOCS-1mRNA in PBMCs of CHB patients was releated to the degree of liver inflammation.
