原发性肝癌组织中长链非编码RNA外泌体复合物7的表达及临床意义

【摘要】:目的探讨长链非编码RNA外泌体复合物7(LncRNA EXOC7)在原发性肝癌组织中的表达及临床意义。方法收集于宣城市中心医院2010年1月-2014年1月行肝癌手术患者的肝癌组织及其癌旁组织标本79对,采用real-time PCR分别检测肝癌组织和癌旁组织中LncRNA EXOC7的表达情况,分析其与病理特征和预后的关系。计量资料2组间比较采用t检验,计数资料2组间比较采用χ2检验或Fisher确切概率法,术后患者的生存分析采用Kaplan-Meier方法,使用Cox模型分析影响预后的危险因素。结果LncRNA EXOC7在肝癌组织中表达水平明显高于癌旁组织(6.211±0.637 vs 2.924±0.415, t=4.106, P<0.01)。肿瘤大小(χ2=5.157, P=0.023)、有无门静脉癌栓(χ2=4.049, P=0.044)、有无脏器转移(χ2=4.345, P=0.037)和TNM分期(χ2=6.479,P=0.011)在LncRNA EXOC7高表达和低表达组间差异均有统计学意义。LncRNA EXOC7高表达组的无瘤生存期(χ2=8.215, P<0.001)和总体生存期(χ2=6.091, P=0.001)较低表达组短。Cox回归分析显示LncRNA EXOC7表达、肿瘤大小、脏器转移和TNM分期是影响原发性肝癌预后的独立因素(P值均<0.05)。结论LncRNA EXOC7参与调节原发性肝癌的发生发展,有望成为一种新的肝癌预后参考指标。
【Abstract】:ObjectiveTo investigate the expression and clinical significance of long non-coding RNA (lncRNA) exocyst complex component 7 (EXOC7) in hepatocellular carcinoma (HCC). MethodsA total of 79 pairs of HCC tissue samples and adjacent tissue samples were collected from the patients who underwent HCC surgery from January 2010 to January 2014. Real-time PCR was used to measure the expression of lncRNA EXOC7 in HCC tissue and adjacent tissue, and its association with pathological features and prognosis was analyzed. The t-test was used for comparison of continuous data between two groups, and the chi-square test or the Fisher’s exact test was used for comparison of categorical data between two groups. The Kaplan-Meier method was used for the analysis of survival and recurrence curves after surgery, and the Cox regression model was used for the analysis of survival and recurrence data. ResultsThe expression of lncRNA EXOC7 in HCC tissue was significantly higher than that in adjacent tissue (6.211±0.637 vs 2.924±0.415, t=4.106, P<0.01). The expression of lncRNA EXOC7 was associated with tumor size (χ2=5.157, P=0.023), presence or absence of portal vein tumor thrombus (χ2=4.049, P=0.044), presence or absence of organ metastasis (χ2=4.345, P=0.037), and TNM stage (χ2=6.479, P=0.011). The high lncRNA EXOC7 expression group had significantly shorter disease-free survival and overall survival than the low lncRNA EXOC7 expression group (χ2=8.215, P<0.001; χ2=6.091, P=0.001). The Cox multivariate regression analysis showed that lncRNA EXOC7 expression, tumor size, portal vein tumor thrombus, organ metastasis, and TNM stage were independent factors for the prognosis of HCC (all P<0.05). ConclusionLncRNA EXOC7 is involved in the regulation of the development and progression of HCC and may become a new reference index for the prognosis of HCC.
